For isolation or analysis of anti-collagen antibodies

Frozen serum from DBA/1 mice with CIA, and serum from control and naïve mice. Shipped frozen on dry ice.

Source of high titer anti-chicken type II collagen or anti-bovine type II collagen mouse antibodies.

Serum will be positive for anti-collagen antibodies in ELISA assays at dilutions of 1:100,000 or more.

Serum may be used to isolate anti-collagen antibodies or as a standard for measurement of anti-collagen antibodies in ELISA assays. To generate the standard curve, we recommend starting at 1:1000 dilution and making 1:3 or 1:5 serial dilutions.

For our own studies at Hooke, we normally test one lot of serum and then freeze many 25 µL aliquots for use as standards in multiple ELISA assays.

Preparation

DBA/1 mice 8 to 12 weeks of age are acclimated to our facility for at least 7 days.

CIA is induced according to the protocol CIA Induction in DBA/1 Mice.

Mice are euthanized after 20 or more days of clinical CIA (typically at 35 to 50 days after immunization). Blood is immediately collected and serum is isolated and frozen at -80 °C.

Serum from multiple mice is pooled and stored at -80 °C until shipment.

Frozen serum, pooled from multiple mice, supplied in a 1.5 mL cryogenic tube on dry ice.

Storage

Keep frozen until use. Storage at -80 °C is recommended.
